摘要:从细胞免疫、体液免疫、非特异性免疫三方面研究富硒青钱柳-蛹虫草复方对C57小鼠免疫功能的影响及抗氧化作用。将C57小鼠随机分为阴性对照组、阳药组、富硒青钱柳-蛹虫草复方低、中、高剂量组。分别进行Con A诱导脾淋巴细胞体外增殖实验、血清溶血素实验、巨噬细胞吞噬鸡红细胞实验及自然杀伤细胞NK细胞活性实验,评价富硒青钱柳-蛹虫草复方对小鼠免疫调节功能的影响。此外,通过流式细胞仪测定不同浓度的富硒青钱柳-蛹虫草复方对NIT-1细胞胞内ROS水平的变化。结果显示,与阴性对照组比较,富硒青钱柳-蛹虫草复方对小鼠的体重、脾脏/体重比值、胸腺/体重比值无显著性影响(P>0.05),低、中、高剂量组能显著促进Con A诱导的淋巴细胞增殖(P<0.05),高剂量组对半数溶血值的增加有显著性影响(P<0.05),且低、中、高各剂量组能显著增加吞噬指数和NK细胞活性(P<0.05或P<0.01)。且样品浓度为25~100 μg/mL时,胞内ROS水平与模型组比较显著降低(P<0.01)。结果表明,富硒青钱柳-蛹虫草复方有效增强机体免疫力,具有显著的抗氧化功效。

Selenium-enriched Cyclocarya paliurus and Cordyceps militaris Compound Immunity and Antioxidant Experiment Research in Mice

Abstract:From the cellular immunity, humoral immunity and nonspecific immune research Se-enriched Cyclocarya paliurus-Cordyceps militaris compound on the effect of C57 mice immune function and antioxidant function. The C57 mice were randomly divided into negative control group, positive drug group, Se-enriched Cyclocarya paliurus-Cordyceps militaris compound low, medium and high dose groups. Con A induced proliferation of spleen lymphocytes in vitro, serum hemolysin, macrophage phagocytosis of chicken red blood cells, and NK cell activity were conducted respectively to evaluate the effect of Se-enriched Cyclocarya paliurus-Cordyceps militaris compound on the immune regulatory function of mice. In addition, the changes of ROS levels in NIT-1 cells with different concentrations of Se-enriched Cyclocarya paliurus-Cordyceps militaris compound were measured by flow cytometry. The results showed that the Se-enriched Cyclocarya paliurus-Cordyceps militaris compound had no significant effect on the body weight, spleen/body weight ratio and thymus/body weight ratio of mice compared with the negative control group (P>0.05), Con A induced lymphocyte proliferation was significantly promoted in low,medium and high dose groups (P<0.05), the high dose group had a significant effect on the increase of half of the hemolysis value (P<0.05). The phagocytosis index and NK cell activity were significantly increased in the low, medium and high dose groups (P<0.05 or P<0.01). And when the sample concentration was 25~100 μg/mL, the intracellular ROS level was significantly lower than that of the model group (P<0.01). The results showed that the Se-enriched Cyclocarya paliurus- Cordyceps militaris compound has the ability to enhance immunity and the antioxidation.
